Dissociation Between Neural Processing and Negative Emotion in Methamphetamine Dependence

Methamphetamine dependence has been associated with mood disturbances, antisocial behavior, cognitive deficits, and abnormalities in brain structure and function that suggest increased reactivity of systems involved in the generation of emotion, compounded by deficits in prefrontal cognitive control systems. A cross-sectional study of healthy adults (n = 20) and methamphetamine dependent adults (n = 18), recruited through advertisements in the greater Los Angeles area, was used to determine if methamphetamine dependence is associated with abnormalities in response to negative emotional stimuli and regulation of negative emotion with cognitive reappraisal. The main outcome measures were self-ratings of emotion intensity and regional brain activity as assessed by functional Magnetic Resonance Imaging contrast during emotion and regulation of emotion. Analysis showed individuals that are methamphetamine dependent demonstrate increased activation of neural systems involved in generating emotion (nucleus accumbens, insula, left lateral orbitofrontal cortex) and a deficit in the activation of neural systems implicated in the cognitive regulation of emotion (left inferior frontal cortex), compared to control subjects. These abnormalities are consistent with the neural dysfunction characteristic of addiction and likely contribute to mood disturbances and antisocial behavior. Further research to identify predisposing factors and to improve therapies that target emotion dysregulation may help prevent the development of drug dependence and improve the response to treatment....
